The agreement, which will be made during a conference call with drug regulators, is a major blow to the US drug industry, with drugmakers and wholesalers accounting for nearly three-quarters of total revenues.
The deal is the result of AstraZeneca's efforts to obtain the patents for Crestor on the drug's label, which was acquired in 2005, after it was developed in collaboration with AstraZeneca.
But the company's patent on the drug expired last month, giving AstraZeneca a significant discount. A similar deal was also struck in 2012 with AstraZeneca's subsidiary, Eli Lilly & Co., which had acquired the drug in 2004.
The acquisition cost AstraZeneca an estimated US$9bn in 2004, a fraction of the total US pharmaceutical industry's sales.
But by the end of 2005 Crestor was available to AstraZeneca for around £1.8bn, the equivalent of about half its sales.
AstraZeneca's strategy to avoid the drug's patents for a time, however, has not been successful, and it faces stiff competition from generic competitors such as Teva Pharmaceutical Industries.
AstraZeneca is the latest drug maker to join the fray, with the firm's UK patent on its cholesterol-lowering drug Crestor already in effect.
In May last year, the US pharmaceutical industry reported a price cut of 5.7% and a loss of over 2% on Crestor, which is also the company's biggest rival.
In the US, Crestor sales had increased to US$9.9 billion in 2005.
AstraZeneca's patent on the cholesterol-lowering drug was valid for more than five years, and it's still in the process of being able to sell Crestor for around £1.8bn.
AstraZeneca's other cholesterol-lowering drugs include Crestor, Zocor and Vytorin, which have been on the market since 1999, and its cholesterol-lowering therapy, Zetia.
Crestor's patent on Crestor expired in May 2004, but it is not yet due to expire.
However, AstraZeneca has filed an appeal with the Food and Drug Administration in March 2005 to allow the drug to be marketed as a generic medicine.
The company, which has the rights to Crestor, says its appeal is over patent litigation, and AstraZeneca is now seeking a judge to overturn that decision.
AstraZeneca and Teva are expected to meet the terms of the appeal in June, and will be asked to pay the court to invalidate the Crestor patent.
AstraZeneca's patent on Crestor expired in March 2005, but it is still allowed to sell it.
AstraZeneca has also been selling a cheaper version of the drug, called Nexium, which it is now developing in combination with Teva.
But a Teva spokeswoman told the BBC: "We do not believe that it is in the best interest of the company, and therefore we will not be making any further payments."
AstraZeneca is currently facing US court cases, though it will likely be heard in May.
In a statement, AstraZeneca said it was "confident that the decision to sell Crestor will be upheld" with no immediate impact on its revenues.
The company also agreed to pay US$4.2bn (£3.2bn) in cash, US$3.2bn (£3.6bn) in future, plus US$1.6bn in interest and fees, and US$2.3bn in royalties on sales of its rights to Crestor.
The Anglo-Swedish drugmaker has also agreed to pay US$3bn in 2024 on a $1.2bn charge to its shareholders and the UK-based drugmaker, Pfizer, will be reimbursed by its shareholders of US$5.8bn.

Crestor, containing rosuvastatin, is a widely prescribed statin medication. It has been a significant player in the management of high triglycerides andbad LDL (a high-saltty-cholesterol athelete’s foot, not cholesterol-related) in both adults and children. In the United States alone, it is estimated that more than 80% of adults and children will develop at least one of these conditions. Crestor is approved for adults over 60 and for children 65 years and over with a low risk of developing a statin-associated condition (AAS/CAD).
Rosuvastatin, like Crestor, can be used by adults and children under the age of 18. It is available in oral tablets, capsules, and liquids. The recommended starting dose of rosuvastatin for adults over 60 is 40 mg once daily, but it may be increased to 80 mg once daily to reduce the likelihood of adverse events.
